POS0384 A NOVEL MECHANISM LINKING MUCOSAL BACTERIA WITH AUTOANTIBODY RESPONSES IN RA: ACETYLATED BACTERIAL LYSATE AS A MODEL ANTIGEN

چکیده

Background: Rheumatoid arthritis (RA) is characterized by autoantibodies against post-translationally modified proteins (AMPA) such as citrullinated, carbamylated and acetylated proteins. Importantly, these antibodies are highly multireactive, they often recognize more than one of post-translational modifications. Despite extensive research, the antigens inducing breach tolerance remain unknown, although microbial suspected. Various bacteria known to be capable acetylation, therefore, it intriguing know what mechanisms can underlie towards development anti-acetylated protein (AAPA). Objectives: To investigate whether bacterial origin (1) recognized human derived AMPA expressing B cells; (2) induce when used immunize mice. Methods: Acetylated E. coli were acquired with two separate methods (Figure 1A): culturing in a condition promoting auto-acetylation (intrinsically proteins, IABP), or directly acetylating lysate-derived via chemical reaction (extrinsically BP, EABP). ovalbumin (AcOVA) served positive control for AAPA induction mice, non-acetylated BP (NABP) phosphate buffer saline (PBS) negative control. Mice immunized resulting antibody response was studied ELISA. Furthermore, EABP/IABP/NABP investigated recognition human-derived ELISA AAPA-expressing cells spleen tyrosine kinase (Syk) phosphorylation assay; fibrinogen native Results: Repetitive immunization mice EABP resulted an recognizing acetylated, citrullinated titers exceeded AcOVA substantially higher NABP-immunized 1B). Human-derived monoclonal IABP (not shown). cell activation (measured Syk phosphorylation) assay indicated that (to lesser extent) IABP, but not NABP 1C). Conclusion: potent cross-reactive responses AAPA. This suggests could possibly involved RA. A tri state mechanism for oxygen release in fish hemoglobin: Using Barbus sharpeyi as a model

Hemoglobin is a porphyrin containing protein with an a2b2 tetrameric structure and like other porphyrin compounds shows spectral behavior of species specific characteristics. Researchers tend to relate bands in the hemoglobin spectra to certain structural and/or functional features. Given the fact that hemoglobin is the main oxygen carrier in animals functioning through the Oxy«Deoxy equilibriu...

Immunopathological evaluation of recombinant mycobacterial antigen Hsp65 expressed in Lactococcus lactis as a novel vaccine candidate

Bovine tuberculosis (TBB) is a zoonotic disease distributed worldwide and is of great importance for public health and the livestock industry. Several experimental vaccines against this disease have been evaluated in recent years, yielding varying results. An example is the Bacillus Calmette-Guérin (BCG) vaccine, which has been used extensively in humans and tested in cattle showing mixed resul...

A novel M cell–specific carbohydrate-targeted mucosal vaccine effectively induces antigen-specific immune responses

Mucosally ingested and inhaled antigens are taken up by membranous or microfold cells (M cells) in the follicle-associated epithelium of Peyer's patches or nasopharynx-associated lymphoid tissue. We established a novel M cell-specific monoclonal antibody (mAb NKM 16-2-4) as a carrier for M cell-targeted mucosal vaccine. mAb NKM 16-2-4 also reacted with the recently discovered villous M cells, b...

Expression of HA1 antigen of H5N1 influenza virus as a potent candidate for vaccine in bacterial system

The impending influenza virus pandemic requires global vaccination to prevent large-scale mortality and morbidity, but traditional influenza virus vaccine production is too slow for rapid responses. In this study, bacterial system has been developed for expression and purification of properly folded HA1 antigen as a rapid response to emerging pandemic strains. Here, a recombinant H5N1 (A/Indone...

Docking and Biological Screening of Bezo[A]phenothiazinones as Novel Inhibitors of Bacterial Peptidogloycan Transpeptidase

Rising cases of antibiotic-resistant bacteria is a public health concern. Many approved antibiotics target penicillin-binding proteins example peptidoglycan transpeptidase (PTPase). Due to wide pharmacological activity of phenothiazines, new styryl, aryl, alkynyl, and thiophenyl benzo[a]phenothiazines were synthesized and their inhibitory potency against PTPasein silico and Gram-po...
